Density = total cells / (fields × field area). Default field area at 40× objective is ~0.159 mm².

How do I measure field area?
Use a stage micrometer or consult the manufacturer's table. At 40× with a 20 mm field number the area is ≈ 0.196 mm²; at 40× with 18 mm FN ≈ 0.159 mm².
How many fields should I count?
At least 5 non-overlapping fields for a rough estimate; 10–20 for a publishable measurement.
