Which chamber does the 10⁴ assume?
A standard Neubauer hemocytometer: each large square is 1 mm × 1 mm × 0.1 mm = 10⁻⁴ mL. So total spores per mL = spores counted × 10⁴ × dilution.
How many squares should I count?
Count at least 4 large squares and average. For precision below 5% coefficient of variation, count 5 or more.
